Research Abstract

We examined about the interaction between gene transcription factors and intracellular signal transduction, using the osteoblast-rich bone cells and osteoblastic Mc3T3-E1 cells(E1 cells)cultured under a low-calcium environment for the prevention of alveolarbone collapse seen in the patients with periodontal disease.
In the bone cells cultured in a low-calcium medium, intracellular calcium content and free calcium concetration significantly decreased, compared with those in normal one (p<0.01)
It was found that the binding sites of NF-kB,CREB and Oct-1 of gene transcription factors existed in the sequence in nuclear DNA in E1 cells. DNA-protein binding activity of NF-kB and Oct-1 seemed to be lower in the cells cultured in a low-calcium medium than in normal one. As there is electrophoretically the differencein the position of band of DNa-protein complex between E1 cells in noumal environment and that in a low-calcium one in CREB,it was shown that DNA-protein complex formation seemed to be disturved in a low-calcium environment in CREB.
From these facts, the inhibitory interaction between the function of gene transcription factors and that of intracellular signal transduction seemed to be closely related with the alveolar bone collapse phenomenon in the patient with peiodontal disease.
