| Research Abstract |
本研究は、潜伏EBV活性化にスイッチとして働くBZLF1遺伝子の発現を制御する細胞因子を同定し、ウイルス活性化機序の解明に資することである。
昨年度の研究で、抗Ig抗体処理、未処理Akata細胞より粗抽出液を得、BZLF1プロモーターの抗Ig抗体応答領域27塩基を含む40塩基を合成し、ゲルシフトアッセイを行った。しかし、特異的なバンドは認められず、昨年度の引き続き種々条件を変えて検討したが、最終的に特異的なバンドを検出することができなかった。
BZLF1プロモーターの解析からは、転写開始点上流-552から-221に負の制御域があり、その下流-203から-177に抗ヒトIg抗体応答に必須な領域がある。この配列のモノマーでは抗Ig抗体にほとんど反応せず、3ないし4個つないで初めて応答した。従って、ゲルシフト法にモノマーの配列を用いたために特異的なバンドが検出されなかった可能性がある。今後、-203から-177の部分を3ないし4個つなげたヌクレオチドをプローブとしてゲルシフトアッセイを行う予定である。
