Research Abstract	本年度は、多くの動物の腸内で最優勢菌であるBacteroidaceneの16S rRNAの遺伝子解析を中心に行い、動物腸内から分離されるBacteroides,Porphyromonas,PrevotellaのGannsについてそれぞれに特異的なプライマーをデザインしてPCR法により簡便に同定できるシステムを作ることと第一の目標に行った。始めに、Genns Becteroiclesについて検討したところ、16S rRNAの薬1500塩基をPCR法により増幅したものにB,idistarsonisとB,vulgatusを除いたBacteroidesについては5′-ATT AAT CTT TCT TTC GAA AGG CTA Tの配列のプローブを用いてハイブリダイゼーションを行うことにより、容易に他の菌群と識別できることが明らかとなった。B.vulgatusについては他の菌種と異なり、反応最度の違いから5′-ATT AAT CTT CCT TTC AGA AGG CTG Tの配列のプローブを用いることでより特異的に検出することができた。また、もともと他のBacteroidesとはクラスターの異なるB.distarsohisは、5′-GGG ATG AAG GTT CTA TGG ATC GTAの配列のプローブを用いて特異的に検出することができた。現在までのところ、この3種のプローブと反応しないものはGenns Bacteraidesからは除外することができると考える。今後さらに、同様の方法でPorphyronplnsとPrevotellaについてクラスターごとのプローブをデザインして、Bacteroidacercの中のGennsの簡易同定法を確立する。
