| Research Abstract |
本年度は、前年度作製したBacteroides(Bac)とB.vulgatus(Vu1)に対するプライマーと本年度デザインしたPorphyromonas(Por),Prenotella(Pre)のGenusに特異的なプライマーを、用いてPCR法により基準株とヒトおよび動物腸内からの分離株、さらにBacteroides subgroup以外の代表的な腸内フローラ菌種での特異性を検討し以下のような結果を得た。
標準株による確認:Becteroides cluster;Bacプローブを用いて確認を行ったところ、B.vulgatus以外のBacteroides clusterに属する株は全てpositiveとなった。また、Vulプローブを用いてB.vulgatusの確認を行ったところ識別可能であった。そこで、両者を混合プローブとして用いることを検討したが、Tm温度の違いによりバックグラウンドが大きくなることから、正確な識別が困難となり、両者別々に用いることとした。
Prevotella custer;Bacteroides cluster,Prevotella clusterの両方の株がハイブリダイズした。ただし、この両者以外はpositiveとなる株はなかったので、BacとPreの両方のプローブを組合わせて用いることで識別可能であった。
Porphyromonas cluster;Porphyromonas clusterとその他のBacteroides subgroup(New1,New2)がハイブリダイズした。しかし、この2種以外はpositiveとならなかったので、Porプローブでハイブリダイズする株はBacteroides cluster、Prevotella cluster以外のBacteroides subgroupと考えられる。
分離株による確認:生物、生化学性状テストにより、Bacteroides cluster、Prevotella cluster、Porphyromonas clusterそれぞれに含まれると同定された分離株を用いて、Bac,Vul,Pre,Por各プローブによる確認を行った。どの菌株も同定したcluster内のプローブとハイブリダイズした。従って、これら分離株でも今回使用したプローブが使用可能であることが明かとなった。
他の代表的な腸内フローラ菌種との確認:すでに同定されているBacteroides subgroup以外の代表的な腸内フローラ菌種(Lactobacillus、Bifidobacterium、大腸菌など)でも同様の検索を行った。Bac,Vul,Pre,Por各プローブともにどの菌株ともハイブリダイズしなかった。
