| Research Abstract |
本研究は、分子進化学的アプローチに基づいて塩基配列を解析し、その遺伝子機能の予測を行うことにある。
本年度は、その基礎となる研究の方法論の確立と、実験的なパイロットスタディを実行した。まず、DNAデータベースが、遺伝子機能予測を容易に行えるような検索ソフト群の開発を行った。また、それらのDNAの塩基配列データから分子系統樹を構築するため、今までにはないMDL原理というアルゴリズムを用いて新しい分子系統樹作成法を開発した。
さらに、塩基配列から遺伝子機能を予測するには、遺伝子が複数の機能的ドメインに分かれていることを確認し、それらの機能的ドメインをどのように同定するかを研究した。特に、セリンプロテアーゼ系の遺伝子に注目し、その機能的ドメインの進化的変遷を解明することにより、機能的ドメインを同定する方法を開発した。
実験的には、主要組織適合性抗原の遺伝子領域に的を絞って、人類集団のHLAクラスIIの対立遺伝子を塩基配列から同定したり、ウズラのMHCクラスI遺伝子の同定を試みたりすることによって、塩基配列から遺伝子機能を予測するためのパイロット研究を行った。
