Research Abstract |
A new strain of Vibrio cholerae O139 Bengal was first discovered in India in 1992 and a recurrence of pandemic of cholera due to the new cholera Vibrio occurred in India since 1996. To prevend pandemics of the diseases development of vaccine is most effective. (1) Department of a candidate live vaccine : A mutant of V.cholera O139 that lacks genes for V.cholera O139 that lacks genes for cholera toxin, zonula occludens toxin, accessory cholera toxin, and Eltor hemolysin. (2) Development of a candidate killed vaccine : V.cholerae O139 was killed by Co 60 irradiation The killed cells moved with flagella and adhered effectively to M cells. (3) For an adjuvant to raise effective mucosal immunity, active center of A submit of cholera toxin was inactivated by replacing responsible amino acids. The mutant cholera toxin with the nontoxic A submit exerted an effective adjuvant activity by stimulating Th1 cells. (4) Genes responsible for synthesizing 0139 LPS were analyzed.
