| Research Abstract |
本研究ではこれまでの我々の研究を発展させ、単一のポリペプドにウイルスの脱外被阻止・複製阻止・細胞間移行・全身移行の阻止・ウイルスタンパク質成熟化阻止などの多機能を持たせる技術を確立し、これによって従来に無いウイルスの属を越えた抵抗性を発現する植物を作出し、マルチスペクトラムなウイルス病防除を可能にする抵抗性遺伝資源の構築を試みることを目的として行った。
まず、Potyvirus科のウイルス(potato virus Y、 azuki bean mosaic virus、 tobacco etch virus)、 Capillovirus属のcitrus tatter leaf virus、およびTrichovirus属のpotato virus TのゲノムをDNAシークエンサーを用いて解析し、polymerase gene(POL)、 cell-to-cell movement protein gene(MP)、 protease gene(PRO)、 coat protein gene(CP)の配列をもとに、それらの遺伝子のdeletionクローンを作製し、MPおよびCPのそれぞれを発現する植物を作出し、抵抗性発現に必要な各タンパク質の最小限の領域を確定した。MPおよびCPにつては、不活性なミュータントを発現する植物の抵抗性について詳細に解析した。
