| Research Abstract |
我々は、血中有核球中のAFP mRNAの有無をnested RT-PCR法により検出し、その有無と臨床経過をprospectiveに追跡し、前年度に報告したように末梢血AFP mRNAの有無が流血中に存在する肝癌細胞の存在を強く示唆することを確認した。今年度はその手法の感度をあげ、腫瘍径の小さい早期肝臓癌患者においても頻度は少ないものの、流血中での腫瘍細胞が検出しうることを明らかにした。また、流血中に腫瘍細胞を放出する癌原発巣の治療により抹消血でのAFP mRNAが陰転化することを示した。今後腹部超音波検査の進歩によってますます径の小さい肝癌の増加が予想されることより、治療の指標としての本マーカーの測定は意義あるものと考えられる。今後は流血中のAFP mRNA数の変化を観察し、抹消血中の腫瘍細胞の定量的考察もすすめていく。
