癌の新しい診断と治療のための磁性体ハイブリッドの開発

Research Abstract

癌の的確な治療を行なうためには、癌の存在診断だけでなく質的な診断、すなわち、癌の浸潤範囲や転移の程度を知ることが極めて重要である。しかし、癌の質的診断を的確に行なうには従来のCTやMRIでは不充分である。本研究の目的は癌と特異的に反応するモノクロナール抗体を癌の存在する部位に特異的に分布させ、従来不可能であった正確な質的診断を手術中に可能とすることである。
(1) 大腸癌と特異的に反応するキメラ化A7-Fab分画をクロラミンT法で^<131>I標識し、抗体活性が保持されていることを確認後にヒト大腸癌細胞とin vitroで反応させて、小型のγ-カウンター(Neoprobe portable gammadetecting probe) (GDP)で検知できることを確認した。
(2) ヒト大腸癌培養細胞株WiDr移植ヌードマウスを用いてin vivoで検討した。^<131>I標識キメラ化A7-Fab分画をマウスの尾静脈から静注し、腫瘍への^<131>I標識キメラ化A7-Fab分画の集積について従来のγ-カウンターによるカウントとGDPのカウントを比較したところ、その結果は相関した。また、GDPで測定したカウントの腫瘍/バックグラウンド比は投与後2日目で3に達し、8日目まで持続した。
(3) モノクロナール抗体A7と磁性鉄を結合させてハイブリッドを作成した。磁性鉄は粒子径が小さいGeorgia-Pacific West社製を用いた。A7とLignosite FMLとをsonicationの後に遠心分離し、A7-Lignosite複合体を作成した。A7-Lignositeの抗体活性はほぼ完全に保持されていた。
以上より、^<131>I標識キメラ化A7-Fab分画を用いて、手術中に癌病巣の認識を正確かつリアルタイムに行なうことが可能となった。また、磁性鉄の応用により、放射線汚染の無い安全な臨床応用が可能であると考えられた。
