Research Abstract	口腔乾燥症(ドライマウス)乾燥性角結膜炎(ドライアイ)などの症状を呈する臓器特異的自己免疫疾患の一つであるシェ-グレン症候群(SS)のEBV再活性化現象の解明とその制御機構を把握することを目的として、Epstein-Barrウイルス(EBV)再活性化マーカーであるZEBRAの発現解析さらにはEBV再活性化に関連する宿主細胞因子の単離を試みた結果、以下に示すようなことが明らかとなった。 1.RT-PCRによる検討からSS患者由来B細胞株SS-BCLにはZEBRA mRNA発現が認められ、末梢血中でもEBV再活性化が検出された。 2.EBV再活性化モデルAKATA細胞を使用したmRNA differential display法よりEBV再活性化状態に特異的に発現している遺伝子断片が得られた。 3.塩基配列解析の結果、mRNA differential display法により得られた遺伝子断片はP/CAF塩基配列と完全一致を示す遺伝子断片であった。 4.degenerate PCRによって一次アミノ酸配列上CREB/ATF、NF-ATファミリーなどのbasic Leucine Zipper構造(b-ZIP)モチーフを有する因子b-ZIPファミリーに類似した構造をもつ遺伝子断片が得られた。以上の結果から、これらの転写因子の結合を阻害することによりEBVの活性化を抑制出来る可能性が示唆された。
