糖鎖認識接着分子、セレクチン郡の機能解明と新世代抗炎症、抗癌転移薬開発への応用

Research Abstract

本研究の目的は、糖鎖認識接着分子であるセレクチンファミリー(E-、L-、P-セレクチン)の糖鎖認識特異性、細胞相互認識機構、その後におこる情報伝達機構などを明らかにし、新世代の抗炎症、抗癌転移薬開発へと応用する実験的基盤を確立する事である。
セレクチンファミリーの糖鎖リガンドとしてシアリルルイスxが知られている。我々はこれに加えて硫酸化糖脂質(スルファチド)が強力なL-セレクチンに対する糖鎖リガンドであることをすでに見いだしている。本年度の成果を以下に述べる。
1)上記以外のシアル酸含有糖鎖、硫酸化糖鎖について各セレクチンに対する認識特性を調べた。さらに硫酸化糖脂質であるスルファチドはL-セレクチンのみならずP-セレクチンに対する強い糖リガンドであるが、E-セレクチンに対しては認識が弱いことを見いだした(1995年度日本生化学会などで発表)。
2)セレクチンファミリー共通の糖鎖リガンドであるシアリルルイスxおよびシアリルルイスaの前駆体である糖脂質、nLc4Cer(Type II骨格)およびLc4Cer(Type I骨格)に対するシアル酸転移酵素(既に研究分担者、西らにより遺伝子がクローニングされた)の特異性を明らかにした(Miyamoto,D.,Nishi,T.,Suzuki,Y.et al.,Biochem.Biophys.Res.Commun.217,852-858,1995)。
2)スルファチドは、新しいセレクチン依存性炎症モデルであるコブラ毒因子投与により惹起される肺炎症を強力に抑制することを見いだした(Mulligan,M.S.,Suzuki,Y.,Ward P.A.et al.,International Immunol.7,1107-1113,1995)。
3)尿路結紮腎炎は新しいL-セレクチン依存性腎炎症モデルであることを見いだした。このモデルラットへスルファチドおよびいくつかの硫酸化糖鎖を投与したところこの腎炎を極めて有効に抑制することを確認した(1995年度の日本腎臓学会、日本脂質生化学会、日本薬学会などで発表)。
4)スルファチドは幼弱Bリンパ球の分化および抗体産生を抑制することを見いだした(1995年度日本免疫学会で発表)。この結果は、Bリンパ球の活性化に必要なTリンパ球との接触にセレクチン-糖鎖リガンド間の結合が必要であることを示唆するものであり、免疫学上極めて重要な結果である。以上、初年度の研究は順調に推移した。
