| Research Abstract |
樹状細胞を含むpassenger leukocyteの拒絶反応に与える影響を解析するために、vascularized bone marrow transplantation後のラットアロ皮膚移植片生着について検討した。
(1)WKAラットの皮膚をFischerラットの背部に移植しFK506を2mg/kg単回投与すると約14日目で拒絶される。(2)皮膚片とともにWKAラット由来後肢(皮膚、骨、筋肉を含む)を吻合にてFischerラットに同時移植すると、皮膚片の生着日数は約32日と著明に延長した。(3)後肢の成分のうち、どの組織が皮膚の生着延長に関与しているのか検討するために、後肢のかわりに、 vascularized skin (VS)、vascularized muscle (VM)、vascularized bone and muscle (VBM)の同時移植を行った。移植皮膚片はVSでは23日、VMでは23日、VBMでは38日であった。したがって骨髄由来passenger leukocyteが皮膚の生着延長をもたらすことが明らかとなった。(4)ドナー由来 passenger leukocyteがレンピエント(♀)に存在することは、ドナー(♂)のY染色体の存在を移植片PCR法にて確認した。
