| Research Abstract |
I.インターカレーションモニタリングPCR(IM-PCR)装置の開発:
1.試作プロトタイプの完成-一応計画通り各パ-ツを調達し,組み合わせてプロトタイプを試作した.パ-ツ間の組合せの不都合も解消できたので,製造承認申請に向け準備に入った.
2.至適条件の確立-PCRの反応条件はプライマーの配列によって微妙に異なり,多分に試行錯誤的であったが,種々の検討の結果,ほぼ統一メニューで実施できる目途が立った.
3.生体試料からの核酸抽出法の確立-AP(凝集分配)型除蛋白剤と半自動プラスミド抽出機の組合せで,血液検体から血球の予備分離なしにDNA/RNAを抽出する方法を確立した。
II.遺伝子検査への適用:
4.C型肝炎ウイルス(HCV)RNA-PCRをRT(逆転写)と組合せることにより,本法がDNAだけでなくRNAにも適用可能なことを実証した.これを実際のHCV感染患者の血清に適用し,他の方法では不可能な10^3コピー以下の微量定量を可能にし、これがインターフェロンの治療効果や肝炎再発の予後判定に極めて重要なことを明らかにした.
5、MRSA(メチシリン耐性黄色ブドウ球菌)の検出-mecA遺伝子を用いて、MRSAとMSSA(感受性菌)の高感度識別が可能なことを示した.
以上の成果により,完全密封型自動・非固相(ホモジニアス)PCR定量装置をほぼ予定通り開発できたものと考える.
