| Research Abstract |
組織や血管内における酸素分圧の測定法は確立されているにもかかわらず、細胞内における酸素濃度測定については推定の域を出ず、直接測定して証明した研究は皆無である。細胞内酸素濃度分布がどのような形で心筋代謝に関与しているのかは、虚血などの病態を解明する上で極めて興味深い。本研究において我々は、最近報告されたphosphorescenceのquenchingの定量化によって組織あるいは溶液中の酸素濃度を測定する方法を単離心筋細胞に応用し、共焦点レーザースキャン顕微鏡下に細胞質内酸素濃度分布を測定するシステムの開発を試みている。また得られたデータを酸素濃度測定下のin vivo収縮性と比較検討し、その意味付けを行うことを目標としている。
