思春期早発症の病態解明

Research Abstract

特に卵巣に於ける卵胞の発育に対してはFSHの役割が重要な位置をしめているので、患者のFSHセプターの構造につき検討し女性における思春期早発症のレセプター異常をスクリーニングした。思春期早発症を診断された患者の血液を採取し、DNAを抽出し、FSHレセプターのイクソンの細胞内領域をコードする部分を増幅出来るようにプライマーを作成した。このプライマーを用いてPCRを行い、理論上正しいと思われるフラグメントをベクターに導入する。複数のクローンのDNAをシークエンスしそのDNA配列を明らかとして、既に我々が報告しているヒトFSHレセプターの配列と比較した。相違部分を確認し、これを基に点突然変異を人工的に作製しCOS細胞に導入しその機能の検討を行った。
一方レセプターの機能調節を検討するためレセプターcDNAを発現ベクターに連結しCHO細胞に導入し恒常的にレセプターを発現する細胞系を確立した。この細胞はFSH添加により用量依存的に細胞内のcAMPの上昇が見られた。この細胞を用いた実験では、FSH前添加によりFSH再添加に対するcAMPにおける反応性の低下がみられた。この結果は思春期早発症において想定されるレセプターの恒常的活性化状態(脱感作機序から逸脱)の機序の検討に使用出来る系と考えられた。さらにこの細胞にフュラ2を前処置した後FSH添加により細胞内のカルシウム上昇を観察した、このことよりヒトFSHレセプターがことなるG蛋白を介してcAMPとカルシウムが情報伝達系として上昇し作動していることが示された。
