| Research Abstract |
PLC_<γ1>、Ash/Grb_2のEGF受容体結合部位のリン酸化ペプチドを合成し、EGF-受容体とPLCδ、Ash/Grb_2の結合への影響を調べた。各々の結合部位の合成ペプチドは特異的にEGF-受容体とPLC_<γ1>、Ash/Grb_2の結合を阻害した。
一方、SH_3に結合するとされるプロリンに富む配列のペプチドRPPPLPPRを合成し、Ash-SH_3とダイナミン、Sosとの結合への効果を調べたところ、このペプチドは両者の結合とも抑制した。これらの結果よりSH_2部位で阻害するペプチドを作ればより特異的にシグナル伝達が阻害でき、SH_3部位で阻害するペプチドを作れば全てのシグナルが抑制できる可能性を示した。
