Research Abstract	本年度の研究においては、導入遺伝子の細胞・組織化学的検出法の確立とモニタリングへの適用を検討した。まず、前者の研究課題においては、導入する標識遺伝子として、抗生物質であるハイグロマイシンの無毒化遺伝子、β-グルクロニダーゼ(GUS)生産遺伝子およびルシフェラーゼ生産遺伝子を用い、これらの遺伝子をベクター内に組み込んだプラスミドを作製し、病原菌類細胞で機能的に発現しうることを明らかにした。初年度の研究において、遺伝子導入法が確立されたので、その遺伝子の発現を多面的に検出するシステムを検討した。検出の方法としては、遺伝子から転写されたmRNAを検出する方法をとり上げ、標識したプローブによる細胞内ハイブリダイゼーションズを行った。この場合、ハイブリダイゼーションに用いるプローブを異なる蛍光色素(ロ-ダミン、フルオレッセイン等)で標識したので、複数のmRNAを同時検出することが可能であった。以上の結果は、本年(1996年)の日本植物病理学会報第62巻576頁から579頁に発表した。また、本法の詳細については、全国農村教育協会から刊行される著書『フザリウム病』において、『遺伝子標識法』として執筆した。一方、モニタリングシステムの構築に関する研究課題においては、病原菌がある種のインドール類縁化合物に対して異なる感受性を示すことを明らかにし、同じく日本植物病理学会報の第62巻414頁から417頁に発表した。
