Research Abstract

In this study, DNA probe method was investigated for rapid identification of algicidal filamentous bacteria isolated from marine environments. Test strains were isolated from sea water, marine algae and marine plants in Kagoshima Bay from 1990 to 1995. The following results were obtained.
1. As GC% of test strains were found to be between 34.6 to 38.9%, they were suggested as to belong to the genera Saprospira, Microscilla or Flexibacter.
2. For the strains isolated in 1990,1991 and 1992, DNA-DNA hybridization was detedted among the strains isolated in the same year while it was not among the strains in the different years.
3. DNA-DNA hybridization was detected between SS92-11 and SS93-7 and among SS95-1 and the strains isolated in 1990, but it was not among SS95-4 and all other strains.
4. On the basis of RFLP patterns for PCR products of 16S rDNA,test strains were classified into three groups such as 90 year group, 91 year and 95 year group, and 92 year and 93 year group.
These results indicate that DNA probe method is useful for rapid identification of algicidal filamentous bacteria.
