ホジキン病病因におけるEpstein-Barr virusの関与-特にスーパー抗原との関連-

| Research Abstract |
(1)未治療ホジキン病患者末梢血中T cellsにおけるV_βレパトワの検索。リンパ腫治療研究会(事務局:金沢大学)を通して、12名の未治療ホジキン病患者末梢血を得ることができた。これらの末梢血よりリンパ球を分離し、T cells中のV_β2,V_β3,V_β5,V_β6,V_β8を発現の頻度をCD4とCD8別々に調べた。このうちの6名はさらにV_β13.6,V_<ββ>17,V_β21.3の頻度も検索した。健常人(福井医大学生)80名と比較したところ、V_β6を除き他のV_βはすべてcontrol群の分布内にきた。V_β6に関しては、2例においては健常人の最大値の2倍以上の増加があり、他の10名に関しては健常人の下限あるいはこれ以下の値をとった。著しい増加をみた1例を寛解後検索したところ、V_β6^+T cellsの頻度は正常域にあった。以上より、ホジキン病においてはスーパー抗原様の物質がV_β6^+T cellsを特異的に活性化しその結果細胞増殖をひきおこし、さらにアポトーシスに至る可能性が考えられる。この現象がホジキン病の病因と関係するのか否かは現在のところ推測の域を出ない。しかし、ホジキン病において、リンパ球の著しい浸潤を見るtypeとの関連が考えられる。
(2)スーパー抗原関与の可能性を検索のため、ホジキン病患者よりEpstein-Barr virusによりtransformしたB cellsの樹立を行っている。一方、V_β6^+T cellsに関しては活性化のマーカーであるCD25,DR,CD69の発現の有無さらに増殖がpolyclonalであるのかoligoclonalであるのかをT cell receptor β鎖のjunctional regionをsequenceして調べている。
