Research Abstract |
Pediatric tumors are characterized by occasional spontaneous regression, as well as their rapid and aggressive growth. We examined this unique feature from the point of view of apoptotic cell death, especially on mass screened neuroblastomas (MS-NBs), whioh have favorable prognoses and even occasionally regress spontaneously. Histological examination of MS-NBs revealed individual cell death and apoptotic bodies, but extracted genomic DNA failed to showtypical ladder-like cleavage. Further, we performed northern blot analysis to assess the expression of high affinity nerve growth factor receptor (TRKA), low affinity nerve growth receptor (LNGFR) and tissue transglutaminase (TS-TG), which are known to be intimately related to apoptosis. MS-NBs were classified into two groups: 1) which show high expression of TRKA and 2) which show high expression of LNGFR and TS-TG. The former seemed to differentiate to nerve cell lineage, whereas the latter cause apoptotic cell death. (Miyagi et al., submitted) We also histopathologically examined pediatric tumors with tendency of spontaneous regression, but failed to obtain evidense of apoptosis. At present, we are planning to evaluate the effects on cell growth, differentiation and cell death caused by transfection of TRKA, LNGFR and TS-TG genes.
