がん細胞の浸潤・転移形質の発現に関与する遺伝子群の検索と同定

Research Abstract

1.BALB/c3T3 A31-1-1ras1000細胞から回収されたヒトの転移遺伝子:
rasでトランスフォームした低転移性のBALB/c3T3 A31に転移能を付与するヒト大腸菌(SW480)由来のゲノムDNAの塩基配列を明らかにした。既知の遺伝子の塩基配列との相同性は現在までのところ認められず,新しい遺伝子である可能性が高い。また、A31-1-1細胞はrasでトランスフォームしても転移性とはならないが、srcでトラッスフォームすると転移性となるという性質をもつので、得られた新しい遺伝子は、がん細胞の転移形質発現を制御するsrcが関与する細胞内シグナル伝達カスケードとの関連が予想される。現在、対応するcDNAと推定されるアミノ酸配列をもとに、この遺伝子産物の機能について検討中である。
2.Neural細胞を用いた転移遺伝子の検索および解析:
マウスの初期発生時期の胎児からのneural crest細胞の分離法を確立した。得られるneural crest細胞は非常に少量(10^3〜)のため、微量細胞からsubtracted expressioncDNA libraryを作成する方法を確立し、cDNAライブラリー作成の準備を終えた。
