| Research Abstract |
本年度はこれまでに作成したLacZ遺伝子を導入したマウスLewis肺癌細胞株(4A1-1)に加えてヒト胃癌の中で肝転移が高頻度で観察される分化型腺癌に類似した転移性のラット分化型前立腺癌細胞株(PLS30LZ)を樹立し,これにlacZ遺伝子を導入して単一細胞レベルで両者の微小転移形成過程を比較検討した.両細胞間で着床後の転移巣の増殖動態には顕著な差は認められなかったが、両細胞の血液中の存在様式や、増殖の場に差異が認められた。Lewis肺癌細胞(未分化癌)は血液中を単一細胞として移行し、着床後早期に血管外に脱出し、管外性増殖を主とするのに対し、PLS30細胞(分化型腺癌)は血液中を大小の細胞魂を形成しながら移行し、着床後血管内で管内性に増殖する傾向があることが明らかとなり、微小転移形成過程が細胞の分化度等に依存して多様である可能性が示唆された。PLS30LZ細胞はヒト胃癌GLM-1株と基本的に類似した転移様式をとるものと考えられ、有用なヒト胃癌の転移モデルになるものと思われる。現在、本株を用いてヘパリン、RGDSペプチド等による転移抑制実験および接着分子受溶体の発現につき検討を進めている。
ヒト胃癌転移性ヌードマウス移植株GLM-1の培養細胞株については樹立を数回試みているがいずれも継代5-6代になると増殖が殆ど停止し、いまだ株化には至っていない。またヒト胃癌非転移株のlacZ遺伝子導入株については発現の安定した株を継続して分離中である。これらに関しては今後なお一層の努力が必要である。
