Research Abstract |
A monoclonal antibody (MoAb), termed 1-7B,which recognized a gut-associated excretory-secretory antigen, was produced by using spleen cells of Schistosoma japonicum infected mouse. We characterized the antigen on a two-dimensional gel electrophoretic pattern (2-D pattern) of proteins extracted from S.Japonicum adult worms. The antigen formed one vertically elongated spot, with an apparent molecular weight of around 100 kDa and an isoelectric point of less than 4. The spot was evident on a 2-D pattern of S.japonicum-infected rabbit serum, indicating that it was a circulating antigen. In addition, because of its lectin binding activity and V8 protease sensitivity, we termed the antigen circulating glyco-proteinaceous antigen (CGPA). Next, a polyclonal rabbit antiserum against CGPA was prepared and used for detection of CGPA on 2-D patterns and paraffin sections of adult S.japonicum and S.mansoni worms. The results showed that CGPA distributed on cytoplasm of gut epithelial cells, only in S.japonicum. Since all five CAs of adult S.japonicum, reported previously by other authours, were known to have immunologically related antigens in adult S.mansoni, CGPA characterized by MoAb 1-7B have not been reported before, on the grounds of differences in their specificity and molecular weight from those in previous reports.
