| Research Abstract |
本プロジェクトに関わる一連の研究において、これまでMφを介する肝障害として報告されてきたPropionibacterium acnes (P.acnes)+LPS誘発肝障害モデルが実はT細胞依存的であることが、ヌードマウスを用いたT細胞移入実験により明らかにされた。また、抗LFA-1抗体を用いた研究から、LFA-1分子を介した免疫担当細胞の接着が肝障害発症に必須であることも証明された。さらに、T細胞依存的な肝障害発症においてはIFN_<-Y>を産生するTh1細胞が重要であり、そのkey cytokineとしてはIL-12が必須であることを初めて明確にした。肝障害発症がTh1-Th2バランスによって制御されていることを世界に先駆け示すことができたのは意義深いと考える。また、Th1-th2バランスの偏りが遺伝的に決定されているマウスにおいて、肝障害の感受性が異なることも初めて明らかにすることができた。
さらに、アレルギーと接着分子に関する研究においては、抗LFA-1抗体の投与によって長期間、IgE産生が抑制されることを証明し、この現象はTh2細胞の免疫寛容誘導に起因することを示した。
肝障害、アレルギーのいずれにおいてもTh1-th2バランスが重要であることが、本プロジェクトの遂行によって明らかにされ、今後はTh1-Th2バランスを支配する遺伝子の追求が重要な課題であると考える。
