| Research Abstract |
1.回虫抗原感作により作成した遅発型喘息モルモットモデルを用いて、漢方製剤である柴朴湯の効果を検討した。柴朴湯は遅発型喘息反応を抑えるとともに、肺組織への肥満細胞/好塩基球や好酸球の集積を抑制した。また、肺組織中のTumor necrosis factor-αやInterleukin-1α、β、Graunlocyte-macrophage colony stimulating factorの発現を免疫組織化学的に検討したところ、柴朴湯の前処理によりれら炎症性サイトカインの発現が抑制された。
2.気管支喘息患者から生検により肺組織を採取し、好酸球の集積に関与する接着因子の発現を検討した。対照疾患と比較して、好酸球,肥満細胞/好塩基球などの炎症細胞上のMac-1,LFA-1,VLA-4の発現が著明に上昇していた。それらのligandであるICAN-1,VCAM-1,E-selectinは血管内皮細胞や気道上皮細胞上に強く発現されていた。
3.ヒト好塩基球性白血病細胞株KU812Fの増殖・分化に対する抗アレルギー剤および漢方薬の効果を検討した。その結果、azelastin,anlexanox,DSCG,baicalein,柴胡サポニンSa/Sd,人参サポニンRh_2、柴朴湯および小青竜湯はKU812Fの分化およびヒスタミン含量を用量依存性に抑制した。
