Research Abstract |
Helicobacter pylori (H.pylori) has causal role of histologic gastritis, gastro-duodenal ulcer and gastric carcinoma. The pathophysiology of histologic gastritis are closely associated with virulence factors of the organism and the host response to H.pylori. In this study, we have investigated the immune response of H.pylori infection. The expression of 66 kD protein was strongly induced in H.pylori after heat shock stimulation. The sequence analysis clearly demonstratedN-terminus of the 66kD protein was identical for HSP 60 family protein. By using rabbit anti-H.pylori HSP antibody, the expression of HSP protein was investigated in H.pylori infected gastric mucosa. Forty H.pylori positive histologic gastritis and 34 gastric ulcer tissues were studied. Eight of 40 gastritis tissues (20.0%) showedHSP expression on the epithelial cells. Eight of 34 gastric ulcer tissues (23.5%) had HSP expression on the epithelial cells. HSP is a cross-reactive protein between H.pylori organisms and H.pylori infected gastric epithelial cells. On the other hand, HSP antibody was detectedin serum of H.pylori infected patients. These results demonstratedthat immune response to HSP was induced in the patients. Although a infiltration of T cells was also observed, those cells had no expression or low expression of gamma/delta T cell receptor. It is required to investigate whether HSP is a target molecule recognized with intraepithelial cytotoxic T cells in H.pylori infected gastric mucosa.
