| Research Abstract |
*本年度は、消化管各部位におけるc-kit発現細胞の細胞学的検索を通して、ペースメーカー細胞の特性、カハールの介在細胞(Interstitial Cells of Cajal;ICC)の細胞学的異型性等を検討するため、免疫組織化学的ならびに電子顕微鏡的に検索を行った。
*材料には、c-kit遺伝子に突然変異をもつWs/Wsラット及び同腹の正常ラット+/+を用い胃、小腸、大腸のICCについて観察、比較した。
*筋層間神経叢の領域にみられるICCは、消化管の部位に拘わらず、共通の微細構造、即ち、豊富なミトコンドリア、大きなgap-junctionの形成、発達した滑面小胞体によって特徴ずけられるが、基底膜、caveolaeは観察されなかった。また、これらの細胞はWs/Wsラットでは観察されなかった。
*一方、小腸の深部筋神経叢では、豊富なミトコンドリア、gap-junctionに加え、基底膜、caveolaeを持つ細胞がみられたが、この細胞はWs/Wsラットでも観察された。胃には深部筋神経叢に相当する神経叢は無く、これに相当するICCも存在しない。
*大腸の筋層下神経叢のICCは小腸深部筋神経叢のICCと似た細胞学的特徴を示し、Ws/Wsラットでも観察された。
*以上の観察結果から、c-kit発現細胞あるいはICCは細胞学的に不均一な細胞群からなり、ペースメーカーとしては筋層間神経叢のICCが第一義的な役割を持つ一方、その他の
部位のICCについては、一層詳細な検索、考察が必要なものと結論した。
