硫酸基糖転移酵素の発現クローニングと癌転移との関連についての研究

Research Abstract

我々の研究室で作製した硫酸化糖脂質を認識する種々のモノクローナル抗体を利用することにより、真核細胞発現ベクターを用いてこれまで困難であった硫酸化糖転移酵素の遺伝子を、発現クローニングし酵素学的な研究と細胞接着物質、細胞間マトリックス、及び血液凝固物質などとの反応性の検討が行えるなどの発展を目指して研究を進めた。
1.真核細胞発現ベクターを用いて作製した、硫酸化糖鎖抗原陽性のHepG2細胞のcDNAライブラリーを、COS細胞にトランスフェクションを行った。硫酸化糖鎖特異的モノクローナル抗体を付着させたプレート上に、トランスフェクタント細胞を反応させ、Hirt法によるplasmid DNAの回収を行った。
この一次Panningを2ク-ル施行した。現在継続中である。
2.末端ガラクトースに硫酸基の結合した構造の中で、最も単純な構造(SM3)の合成に関与する、糖脂質特異的硫酸基糖転移酵素が先日報告されたが、そのcDNAのcoding regionの断片を用いて、硫酸化糖脂質の存在が証明された。(Hiraiwa,N.,et al. Cancer Res. 50:2917-2928,1990)ヒト肝癌細胞株、胆管癌および硫酸化糖脂質の合成を認められなかったヒト正常肝由来細胞の同酵素の転写発現をNorthern blotで調べたところ、同糖脂質合成量と遺伝子転写発現量とは正の相関を見た。現在、同酵素遺伝子を正常肝由来細胞に安定発現させ、硫酸化糖脂質の合成、細胞接着などの機能変化を研究している。
