| Research Abstract |
(1)Aβペプチド末端、修飾特異抗体の作製と特異性の検定
ΑβのC末端(Aβ40,42)を特異的に認識するポリクローナル、モノクローナル抗体はすでに得られ、その特異性は2抗体法ELISAやイムノブロットで確認された。N末端側についても、βアミロイド中に存在が確認されているAβL-Aspl,pyroGlu3断端に対する抗体を樹立した。これに加えて、L-isoAsp(1,7)を含むペプチド(Aβ1-16)を合成し、これに対するモノクローナル抗体を樹立した。
(2)Αβ部位特異抗体を用いたAD脳、Down症脳の免疫細胞化学
AD,Down症剖検例大脳皮質のパラフィン包埋標本から連続切片を作製し、上記の数種類の抗体を組み合わせて連続切片上で免疫染色を施し、各種のAβ蓄積物がどのような性状のAβペプチドを含むかを検討し、あわせて計量的解析も行った。
β蛋白の修飾状態を検討するための特異抗体はほぼ計画通り樹立され、これによりアルツハイマー脳、ダウン症脳組織を系統的に検索することができた。検討を進める間に、組織の固定状態によりβ蛋白アミノ末端側の免疫反応性が大きく影響を受けることが明らかとなった。これに対しては共同研究者のTrojanowski,Lee教授から供与されたアルコール固定パラフィン包埋標本を用いて再検討を行っている。この結果を待って、脳より抽出したβアミロイド蛋白の生化学的解析に着手できる見通しである。
