| Research Abstract |
本研究の目的はglucocorticoidによる中枢性血圧調節機構を検討することである。当初の研究計画に基づいて無麻酔無拘束ラットを用いてデキサメサゾン慢性投与の24時間血圧に及ぼす影響を検討し、併せてtail-cuff法による血圧値との比較を行った。その結果以下のことが明らかになった。
1)Tail-cuff法によるとデキサメサゾン(dexa)投与群では対照群と比較しdexa投与開始24時間後から血圧上昇が認められた。
2)その後dexa投与群では血圧が漸増し7日目にプラトーに達した。このとき二群間の血圧壁は30mmHgであった。
3)dexa投与跡10日目直接法にて24時間血圧をモニターした結果、dexa投与群では対照群と比較し測定の全経過を通じて15mmHgの血圧上昇が認められた。従って、dexa投与下での血圧上昇はtail-cuff法による方が直接法によるよりも大きな値を示した。
4)dexa投与群でも血圧の日内リズムは保たれており、慢性的glucocorticoid投与が血圧の日内リズムに及ぼす影響はヒトとラットで異なることが示唆された。
本研究は当初の計画通り進行し上記のような知見が得られた。本研究の成果は無麻酔無拘束ラットにおいてデキサメサゾン慢性投与が24時間血圧プロフィールに及ぼす影響を検討した世界で初めての報告であり、これらの成果の一部は国際誌に掲載された。従って、本研究は着実に成果をあげており、国内のみならず国際的にも高い評価が得られていると考えられる。
