Research Abstract |
Breast-feeding is the major factor of vertical transmissions during the first year of life in countries where the majority of women were seropositive to human cytomegalovirus (CMV) and breast-feed their infants. Studies in our and other laboratories provided compelling evidence that virolactia was more common in breast milk samples collected during 1 week to 3 months postpartum than in those collected during the first week. We collected colostrum and breast milk from 65 human anti-CMV lgG antibody-positive mothers who breast-fed at 3 days and a month after delivery. When investigated by indirect immunofluorescence assay (IFA), liquid supernatant of colostrum without cytotoxicity was not found to exert inhibitory effect on HCMV-infected MRC-5 cells. The activities of tumor necrosis factor (TNF)-alpha were significantly higher in HCMV DVA-negative colostrum than DNA-positive breast milk (p<0.05). Serum levels of cell free soluble interleukin-2 receptor (slL-2R) of mothers of DNA-positive milk at one month after delivery were significantly higher than those of mothers of DNA-negative milk (p<0.05). It is likely that levels of cytokines such as slL-2R in serum are related to reactivation of HCMV which occurs lo cally in the mammary gland of the lactating mother after delivery.
