Research Abstract |
GTP-binding proteins regulate both exocytotic and endocytotic steps of GLUT4-containing vesicles in rat adipocytes. In the present study, we have shown that a synthetic peptide corresponding to the Rab4 carboxy-terminal domain inhibits insulin-induced GLUT4 translocation, indicating that Rab4 may be essential for the insulin action. Furthemore, insulin stimulated guanine nucleotide exchange on Rab4. This insulin action was markedly attenuated with phosphatidylinositol (Pl) 3-kinase inhibitors, wortmannin or LY294002, suggesting that Pl3-kinase may be an essential signaling component for insulin activation of Rab4.
