| Research Abstract |
1、株化細胞における制癌剤によるアポプト-ジスの誘導(in vitro)
株化胃癌細胞における制癌剤によるアポプト-ジスの誘導に関して、癌細胞の断片化を指標に検討を加えた。その結果、シスプラチン、アドリアマイシンにより、その添加濃度、接触時間に応じてアポプト-ジスが誘導される事が確認された。重要な事は、アポプト-ジス誘導の程度が細胞間で異なり、感受性が一様でない事である。
2、術前化学療法によるアポプト-ジスの誘導(in vivo)
術前に5-FUによる化学療法がなされた胃癌症例、大腸癌症例におけるアポプト-ジスの誘導を調べ、それとc-myc,Ki-67,p53発現との関連を検討した。術前化学療法により胃癌細胞、大腸癌細胞に高率にアポプト-ジスが誘導される事が確認された。しかし、このアポプト-ジスとKi-67,p53など他因子の発現には直接の関連はみられなかった。
3、術前化学療法によるアポプト-ジスの誘導と術後生存期間との関連
胃癌症例を対象に、術前に5-FUによる化学療法を施行し、アポプト-ジス誘導の程度が術後の再発と生存期間に及ぼす影響について検討中である。更に、再発癌、切除不能例の化学療法において、従来からのintensive chemotherapyからmildな、主としてアポプト-ジス誘導をめざした化学療法に変えて、生存期間への影響を検討中である。
