癌抗原遺伝子のコードする蛋白刺激による頭頸部癌細胞傷害性T細胞の誘導

Research Abstract

HLA-A2,HLA-A24の保有者の有無はそれぞれに特異的なプライマーを3つずつペアで合成し、PCRを行うことにより鑑別できるようになった。MAGE-41のcDNAを組み込んだfusion proteinを産生するEcoli(DH5α株)を照射により不活化し、これをstimolatorとし、CTLの誘導を試みたがHLAクラスI拘束性を示さず、CTLの認識するエプトープの同定は無理と判断した。
そこで次のステップとして、HLA-A2と共に発現される癌抗原ペプチドを4種類合成し、IL-4,GM-CSFで誘導した樹状細胞に、これらのペプチドをパルスして抗原提示細胞とし、HLA-A2保有者のリンパ球から、CTLの誘導を試みている。
