Research Abstract

The purpose of this study is to develop multidisciplinary treatments in ocular tumors, using electrochemotherapy, photodynamic therapy and hyperthermic treatment.
Malignant melanoma cells were cultured. Some of these sells were exposed to various intensities of electric pulse alone (500-2500)V/cm). Other cells were then exposed for 30 min to an antiproliferative agent : bleomycin, mitomycin C or 5-fluorouracil with or without electric pulse. Cell proliferation was assessed by cell counting on day 3 and by a H^3-thymidine uptake assay. A significant reduction in the cell number was observed only at 2500V/cm. Bleomycin, mitomycin C and 5-fluorouracil treatment inhibited cell proliferation in a dose-dependent manner either with or without electric pulse. Electric pulse treatment induced an inhibitory effect of bleomycin cell proliferation which was 100 times more prominent than bleomycin alone..
Malignant melanoma cells cultured were transplanted into the eyelid of the mouse. After the growth of the transplanted tumor cells, electrochemotherapy using electroporation method was performed. The drugs used were bleomycin, mitomycin C and 5-fluorauracil. The most effective drug is bleomycin for this electrochemotherapy.
The electrochemotherapy may therefore reduce the necessary dose of antiproliferative agents for the treatment against malignant tumors. We developed this electrochemotherapy for glaucoma surgery.
