Research Abstract

There exists many kinds of hormone, placenta derived proteins and cytokines in placenta tissue. From this stand point of view, it is supposed to exist bone forming properties in placenta tissue. In this research work, bone formation ability from placenta tissue was examined by rat placenta tissue transplantation into mouse thigh muscle tissue and subperiosteum transplantation of calvaria by means of histological and immunohistochemical methods. The placenta tissue employed for transplantation was proved to be abundant for various extracellular matrix components.
In placenta tissue transplantation into thigh muscle tissue, placenta tissue was existed as an amorphous mass, and accumulation of extracellular matrix (ECM) components were negative except tenascin on 2 weeks post operation. On 4 to 6 weeks post operation, radio-opaque mass could be seen in the implanted area. The calcium was gradually deposited in the amorphous placenta tissue and large amount of calcium deposition could be found on 26 weeks post operation. S-100 protein positive cells were existed in the fibrous tissue between implanted placenta tissue and thigh muscle tissue.
In placenta tissue implantation into subperiosteum of calvaria, ECM components could be found in the fibrous tissue located between calvaria and placenta tissue which changed to amorphous structure and showed negative accumulation of ECM componets on 2 weeks after operation. On 6 weeks, hyaline structure was existed in the fibrous connective tissues, and bone tissue was found on 8 weeks.
From these results, placenta tissue contains predominantly ECM components such as glycosaminoglycans, collagens, laminin, fibronectin and tenascin, calcium signaling substances and bone inducing properties such as transforming growth factor beta and bone morphogenetic proteins. Placenta tissue may act firstly calcium signaling from neighboring connective tissue and provided reasonable condition for ossification, and finally forms osseous tissue.
