| Research Abstract |
紫外線照射直後の培養ヒト細胞で活性増大を示し,照射後の多面的細胞応答を制御するプロテアーゼを求めている。これまでの研究成果をふまえて,モデル細胞を用い,直接プロテアーゼ蛋白質を分離抽出するための方策とプロテアーゼ蛋白質の構造遺伝子が活性制御因子の遺伝子を探索するための方策を検討した。前者では目的プロテアーゼがアンチパイン高感受性であることから,アンチパイン結合タンパク質を分離抽出する方法を確立した。後者では,目的プロテアーゼ活性の紫外線によるレベル上昇がある細胞とない細胞を同一系統株として樹立してあるので,Differential display法により候補遺伝子断片を複数個得ることに成功した。この両面から,目的プロテアーゼを求める方策が一応確立できたと考える。
