X線顕微鏡による細胞内構造および物質輸送の解析

Research Abstract	生きたままの状態で、生体分子の動く様子を直接観察する方法として、X線顕微鏡の開発を行っている。これまで、50nmの分解能でウエットな生物試料を観察できる、ゾーンプレートを用いたX線顕微鏡をUVSOR実験施設で完成させた。次に、試料まわりを大気中に出した設計のX線顕微鏡の開発に入り、UVSOR研究施設での共同実験で大気圧中に設置したケイソウなどのドライおよびウエットな生物試料の観察に成功した。現在は、このシステムを立命館大学小型放射光施設への移築が完了し、電子顕微鏡などの観察ツールと同様にルーチンワークができる状態となっている。重金属ラベル等の方法を用いて、細胞内の特定部位や特定分子を観察する方法の開発しをめざし、本システムで観察をおこなった。シナプス小胞に存在するシナプストフィジンを染色したサンプルなどをX線顕微鏡で観察し、糸球体のより細かい構造を見ることができた。
