ヒト表皮膜骨格蛋白質(4.1蛋白質)の機能に関する形態的および生化学的解析

Research Abstract

膜骨格蛋白質についての生化学的研究の進歩と膜機能の定量的測定法の開発により,膜骨格の構造と機能の密接な関わりが明らかになってきた。とりわけ,スペクトリン,4.1蛋白質及びアクチン等に量的,質的異常を認める赤血球では変形能,膜安定性等の機能が低下しており早期に網内系細胞に捕らえられ、寿命が短縮して溶血性貧血となることが解明されている。本研究ではこれらの膜骨格蛋白質の皮膚(表皮細胞)における機能を解明することを目的として以下の検討を行った。
我々は角化異常や細胞増殖をきたしている尋常性乾癬における膜骨格蛋白質の存在・分布について検討を行った結果,乾癬表皮細胞において正常表皮細胞には認められない45kDaのバンドがイムノブロット法で認められ,また乾癬表皮細胞において4.1蛋白質の異常分布を確認した。一方,乾癬表皮においてスペクトリンの分布は正常表皮と特に差は認められなかった(Shimizu et al,Histochem Cell Biol,1995)。一方r30kDa(4.1蛋白質の30kDaドメイン)から作った抗体を用いた乾癬表皮細胞の分布は正常表皮と特に差は認められなかった。
培養ヒト表皮細胞を用いた実験において,Caスイッチにより4.1蛋白質およびフォドリンの細胞質から細胞膜への分布の変化が認められた。さらにフォルボールエステルの添加により同様の分布の移行が認められた。これらの蛋白質の表皮細胞間接着への関与が示唆される(Shimizu et al,Biol Cell,in Press)。
