Research Abstract	実験胃潰瘍研究においては胃粘膜再生治癒の分子機構の解明は最も重要な研究課題の一つである。成長因子様の作用を有するPTHrPはEGFなど他の成長因子と同様に粘膜再生に関わり、治癒促進のmodulaterとして重要の役割を担っていることが推察されるが、生理的な役割は殆ど解明されていない。本研究では粘膜再生、細胞回転にPTHrPの関わる分子機構の解明を目的としている。潰瘍治癒過程におけるPTHrPの発現は免疫組織化学、ISH、ノーザンブロットで観察しsense-、antisense-oligoPTHrPや発現ベクターの組織内導入は計画どうり実施できた。酢酸潰瘍を作成し1週から4週までの治癒期の潰瘍辺縁組織を採取し、パラフィン包埋標本作成とRNA抽出を行った。免疫染色でPTHrPの発現は潰瘍辺縁の再生上皮と肉芽組織内リンパ球に認められ、ISHでもPTHrPmRNAの発現同様の分布を示しPCNA陽性細胞との共存傾向も認められた。ノーザンブロットでは初期の1-2週目では胃壁全層でむしろPTHrPmRNAの発現は低下していたが、これはPTHrPの局在が本来固有筋層に多く発現しているため潰瘍化による筋層欠損を単に反映してとも解釈できる。ISHでPTHrPmRNAの発現が再生粘膜に観察されていることから、この点に関しては今後定量的には粘膜層のみのサンプリングが必要。またsense-oligoPTHrPとantisense-oligoPHTrPをそれぞれの発現ベクターとともに経口投与し潰瘍治癒過程を観察したが、sense-oligoPTHrPは明らかに治癒を遷延させた。このことからPTHrPの過剰発現は再生化を遅延させていることが示唆された。 PTHrPはPCNA陽性再生上皮細胞との相関が高くかつsense-oligoPTHrPの導入が粘膜再生化を遅延させることから、PTHrPの発現の意義は再生粘膜の増殖抑制あるいは過剰増殖の制御のために発現していると考えられる。
