Research Abstract	副甲状腺ホルモン関連ペプチド(PTHrP)は、動脈平滑筋増殖な軟骨成長調節など生理的或るいは反応性の細菌増殖に重要な役割を演じていることを報告してきた。近年実験胃潰瘍研究においては胃粘膜再生治癒の分子機構の解明は最も重要な研究課題の一つである。成長因子様の作用を有するPTHrPはEGFなど他の成長因子と同様に粘膜再生に関わり、健全な治癒促進のmodulaterとして重要の役割を担っていることが推察される。本研究では粘膜再生、細胞回転にPTHrPの関わる分子機構の解明を目的としている。酢酸潰瘍を作成し1週から4週までの治癒期の潰瘍辺縁組織を採取し、パラフィン包埋標本作成とRNA抽出を行った。免疫染色でPTHrPの発現は潰瘍辺縁の再生上皮と肉芽組織内リンパ球に認められ、ISHでもPTHrPmRNAの発現同様の分布を示しPCNA陽性細胞との共存傾向も認められた。ノーザンブロットでは予想に反して初期の1-2週目ではむしろPTHrPmRNAの発現は低下していたが、PTHrPリセプターの発現は逆に増強していた。PTHrPの転写促進因子として知られているEGFやTGFβの発現とPTHrPは治癒初期には連動していないことも明らかになった。これらのことから早期の潰瘍再生過程においてはPTHrPリセプターの発現亢進が重要であり、PTHrPはその後の再生上皮の分化制御に関わっていることが示唆された。またsense-oligo PTHrPとantisense-o ligo PTHrPをそれぞれの発現ベクターとともに経口投与し潰瘍治癒過程を観察したが、sense-oligo PTHrPは明らかに治癒を遷延させた。このことからPTHrPの過剰発現がPTHrPリセプターのdown regulationを引き起こし再生化を遅延させていることが示唆された。今後はPTHrPリセプターの発現を主眼において研究を進め再生治癒に関わるPTHrPおよびPTHrPリセプターの役割について解明したい。
