Research Abstract |
Purpose : In order to carry out the mechanism of bone metastasis of breast cancer, we studied the relationship between breast cancer cells and bone-cell groups. Materials and Methods : The influence of some materials derived from the conditioned medium of breast cancer cell lines (MCF-7, ZR-75, BT-20), which were cultured in our department, to bone- cells were examined. The influence to bone was gained from the examination that ionized calcium of the conditioned medium from mixed cultured breast cancer cells and bone cells derived from rat- femoral bone were measured. Furthermore, an osteoblast extracted from bone marrow's liquid of the patient who were performed bone marrow autoplantation. The apoptosis, induction of bcl-2 and overexpression of p53 oncoprotein were also studied using breast cancer cell lines. re were no significance of ionized calcium levels between conditioned medium of these co-culture and that of control (without bone cells). 2) In the culture of bone marrow cell, we tried to culture bone marrow cells obtained from the marrow cells at the time of the autopsy and small fragments of bone extracted from the gauze of the filter during extracted bone marrow liquid of the patient who were performed bone marrow autoplantation. But stable culture of these bone cells was impossible. 3) In the breast cancer cell lines (MCF-7, ZR-75, and KK, established by our department), the apoptosis and suppression of bcl-2 expression were shown induced by tamoxifen. 4) The apoptosis and the over-expressions of apoptosis-related bcl-2 and p53 oncoprotein were the correlation with the carcinogenesis and progression of breast carcinomas. Conclusively, we could not get the results for mechanism of bone metastasis by breast cancer cell lines, but it was found that apoptosis is caused by Tamoxifen in breast carcinoma cells. Less
