| Research Abstract |
げっ歯類の顆粒性間膜腺(GMG)細胞,子宮NK細胞は流産に関連すると示唆されている。そこで、MHC class II抗原が不適合の交配およびNK細胞欠損のTgE26マウスの自然流産に着目した。MHC クラスII抗原不適合による自然流産モデルとして、CBA/2J(H-2^d)の雄を用いた時の妊娠10〜16日の流産率は、24%であった。perforin陽性、asialo GM1陽性のGMG細胞は、流産胎盤においても間膜腺および基底脱落膜に局在し、胎盤迷路部や胎子遺残物に侵襲している像は観察されなかった。その細胞密度は正常と有意差はなく、さらに、perforinは依然細胞内に含有されており、流産に際して分泌された形跡はなかった。透過電顕による解析でも、流産と正常との間でGMG細胞の形態に差異は見られなかった。さらに、免疫電顕による顆粒内のperforin検出も有意差は認められなかった。一方、TgE26では、胎盤は対照の半分サイズしかなく、間膜腺の形成が認められなかった。さらに、GM1陽性のGM細胞はほとんど存在せず(対照の3%)、小器官および顆粒は著しく未発達であった。GMG細胞は血管周囲に集合する傾向を見せ、子宮細動脈の中膜は肥厚していた。以上の結果から、GMG細胞が流産を誘起させる可能性はないものと示唆される。CBA(♀)×DBA(♂)系において流産部位と正常な部位で差違が見られず、その一方でTgE26がGMG細胞を欠くと同時に高率に流産を起こしたことから、GMG細胞の存在は自然流産を誘起するものではなく、逆にGMG細胞の欠損が自然流産の誘起に関与することが示唆された。
