活性酸素の代謝制御と脳細胞選択死の分子機構解明

Research Abstract

虚血性脳血管障害における海馬の神経細胞死や家族性筋萎縮側索硬化症(FALS)の運動神経障害にスーパーオキシドラジカルやNOをはじめとする活性酸素の関与が示唆されているが、その実態は尚不明である。本研究は、スーパーオキシドラジカルを部位特異的に消去するミサイル機能含有SOD誘導体を開発し、各種脳病態における活性酸素の役割を分子レベルで解析すると同時に、脳血管の加齢変化に共役する神経病変を阻止軽減する予防治療法開発のための理論基盤を確立することを目的として行っている。東北のFALS患者のCu/Zn-SODの遺伝子解析及びSODの三次元構造解析を行い、Exson4に欧米の患者とは異なる変異(Asn84・・,Ser84)が存在すること、及び本部位が正常酵素分子表面では疎水性カチオン壁を形成することを見いだした。SODの三次構造解析から、Exson-4でコードされる部位は酵素Subunit間および分子表面で疎水性カチオンの壁を形成していることが判明した。本部位と特異的に相互作用するアニオン性物質が血管の加齢変化に伴い増加すれば、神経細胞障害の高発症年齢などを説明しうる可能性が考えられる。スーパーオキシドアニオンラジカルの基質である酸素分子はNOと拡散律速で反応するので、その機能は局所の酸素分圧とスーパーオキシドによる左右され、これが脳血管神経病態を規定している可能性が高い。虚血部位ではNOの作用が著しく増強し、活性酸素間のバランスが崩れ、神経細胞のエネルギー代謝が障害されることが判明した。したがって、脳血管の加齢に伴い、活性酸素が神経系障害因子としての性質を増強する可能性が示唆される。
