| Research Abstract |
われわれは世界で初めてEBV増殖に許容性の細胞を分離し、EBVベクターによる遺伝子導入法を確立した。
本研究の目的はこの新たに確立したEBVベクター系を用いてHIV特異的CTLを誘導し、エイズに対する特異的免疫療法を開発することである。今年度は以下の研究を行った。
EBウイルスのEco-RI-B断片の中心部分へバクテリオファージP1.loxP配列、ネオマイシン抵抗性遺伝子を持つ組み換え用プラスミドを作製した。次いで本プラスミドをEBV要請Akata細胞へトランスフェクトし、標的部分で相同組み換えの起こった細胞クローンを分離した。得られた細胞クローンが組み換えEBVを産生することをサザーンブロット法で確認した。
この組み換えウイルスCreをは一過性に発現させることにより、loxP配列を持つ外来DNAを容易にloxP部位へ挿入することができる。現在、感栄性粒子を産生しないようなHIVプロウイルス変異体、gpt遺伝子及びloxP配列を含むプラスミドを作製し、loxP組み換えEBVを持つAkata細胞へトランスフェクトし、HIV組み込みEBVを作整中である。
