Research Abstract	アゲハの感覚器官を調べるためには、まず大量のアゲハを飼育しなければならない。そのため、カラタチなどの食草を確保し、アゲハを採集し産卵させ、飼育につとめた。その結果約600個のサナギを得ることができた。そのサナギを羽化させた後、アゲハのメスのみ300羽から味覚器官である前肢フ節を切り取り、それからRNAを抽出した。前肢フ節と前肢腿節を切り取り、グアニジン塩酸存在下でホモジェナイズしたのちに抽出液をフェノール処理してRNAを分離した。約180μgの全RNAが得られた。これからmRNAを分画する予定である。mRNAをcDNAクローン化し、めざすミカン産卵誘起物質レセプターcDNAを求めるスクリーニングを試みる。そのさい、何をindexとするかが問題となる。まずは、ハエで知られている味覚レセプターの細胞膜貫通部分の塩基配列を用いてスクリーニングをおこなう予定でいる。そのほかとしては、ハエの前肢フ節の甘味感覚レセプターの探究が進んでいる。前肢フ節cDNAライブラリーから細胞膜7回貫通アミノ酸配列構造をプローブとしてクローニングした結果、いくつかのクローンが得られたので、その配列を決定し、そのどれがグルコースレセプターであるかを検討中である。困難なのは、その検定法である。目指すcDNAを培養細胞DNAに組み換え、その発現を確かめたのち、グルコースに対する応答をチェックしなければならないからである。
