Research Abstract	本研究の目的は、種々の動物における形成中心(両生類のオ-ガナイザー相当)の出現の原理と、それ以後の胚発生、とりわけ三胚葉体制の確立における作用という、二〇世紀における生物学上の最大の問題を分子生物学的に、また、比較発生学的視点から開明することを目的とした。この研究はまた、地球上における生命の存在形態や発生様式の多様性が、系統発生と深く関わっていることを、形成中心の分子的機構という具体的なデータに基づき総合的に理解することを目指した。平成8年度においては、各班員が十分な議論をし、さらに研究の情報を交換するために班会議を開催した。この班会議は、「形成中心フォーラム」として開催され班員以外の研究者も多数参加した。この会議で報告された研究は以下のように要約できる。(1)ウニやヒトデのような無脊椎動物では形成中心という概念そのものの意味付けが問われた。これらの動物では原腸形成が植物極の因子によることが推定されてはいるが、その本質は明らかではない。ただ外腸胚形成ペプチドが表皮成長因子のホモログであることなど、その分子的性質の解明が端緒についてことが報告された。(2)原索動物では、いわゆるオ-ガナイザー相当領域で発現する遺伝子が明らかにされ、脊椎動物との相同性が論じられるようになった。しかし遺伝子の相同性のみで形成中心を定義することに対する疑問も指摘された。このような研究は脊椎動物の形成中心の起源を明らかにする上で極めて重要である。(3)魚類や両生類では形成中心、いわゆるオ-ガナイザーが明瞭である。ここではすでに形成中心の機能に関わる遺伝子が相次いで確認され、その分子的作用に関する研究にと進みつつある。一方で、従来から行われてきた実験発生学的な解析も特に鳥類を中心に進められ、胚葉の分化に関わる遺伝子レベルの研究の進展が報告された。以上のように、本研究班の平成8年度の研究は、それぞれの動物群の形成中心のもつ意義とその分子的作用の解析の点で、多くの成果を上げることができた。
