膵癌の発生・増殖と進展及びその制御に関する基礎的研究

Research Abstract	平成11年度にハムスター膵癌実験系を用いて行った結果と進行状況について報告する。 1.平成9年度と10年度に見出されたTrp-P-1の膵管発癌に対するイニシエイションとプロモーション作用の知見をもとに,0.01%,0.02%Trp-P-1を経口投与し長期発癌実験を実施中で,現在実験開始後70週を経過している. 2.短期膵癌発生系における膵管上皮細胞のBrdu標識率を指標に,4.8-DimeIQXのイニシエイション活性を検索した結果,膵管上皮細胞のBrdu標識率の有意な上昇は見られず,イニシエイション活性はないと考えられた. 3.短期膵癌発生系におけるβ-カテニンの関与は否定された. 4.短期膵癌発生系において,0.1%トラネキサムサンは,動物1匹当たりの膵癌の発生個数を抑制した. 5.4-hydroxyphenyl retinamideとaurapteneは,それぞれ10μM,50μMの濃度でハムスター膵癌培養細胞にアポトーシスを誘導した.
