Research Abstract

免疫抑制剤の進歩により、臓器移植の成績は飛躍的に向上してきている。それに伴い、臓器移植の適用の範囲も、驚異的に拡大し、急激なドナー不足の状態が欧米を中心に実現している。そのため、動物の臓器をヒトに移植する異種臓器移植が注目を集めている。ブタをドナーとする異種移植で発生する、超急性拒絶反応を抑制するために、2つの手段が考えられている。第1はヒト補体制御因子(DAF,HRF20)遺伝子の導入であり、第2は、主要異種抗原GALαGALを誘導する酵素α(1.3)galactosyl transferase(α1.3GT)の発現抑制である。α1.3GT遺伝子の発現を抑制する手法として、(1)α1.3GT遺伝子のノックアウト、(2)α(1.2)frcosyltransferase(α1.2FT)遺伝子の導入が考えられる。我々は、マウスembryonic stem(ES)細胞を用いて、マウスα1.3GT遺伝子をノックアウトするためのノックアウトベクターを作成し、このベクターをelectroporarion法でマウスES細胞に導入した。このES細胞をG418を用いて選択し、サザンブロット、ノザンブロット、FACSにてα1.3GT遺伝子の発現を解析した。このα1.3GTノックアウトマウスES細胞では、GALαGAL抗原の発現が抑制され、しかもヒトIga及びIgMを結合の抑制されることが確認できた。これらの結果をもとに、現在ノックアウトマウスを作成中である。α1.2FT遺伝子をブタ受精卵に遺伝子導入しトランスジェニックブタ作成を行った。この遺伝子導入ブタの白血球及び上皮細胞を用いてα1.2FTの発現をRTPCR及び蛍光免疫染色にて確認した。トランスジェニックブタにおいてα1.2FT遺伝子の発現の確認に成功した。現時点ではα1.2FT遺伝子導入トランスジェニックブタの発育及び成長に与える影響が検討されている。ブタα1.3GT遺伝子は最近欧米でクローニングされた。我々はこの結果をもとにしてブタα1.3GT遺伝子の詳細な解析を行った。現時点ではブタα1.3GTの発現において4種類のalternative splicingが確認されている。またブタα1.3GTのcatalytic domainはエクソン9にあることが確認された。我々はこの結果をもとにブタα1.3GT遺伝子をノックアウトするノックアウトベクターを作成した。次にα1.3GTの発現を抑制し、しかもブタα1.2FTの遺伝子を導入する最も理想的な方法として近年注目を集めているα1.2FT遺伝子ノックインベクターを作成した。このベクターは従来用いられているノックアウトベクターのselection markerネオマイシン耐性遺伝子の代わりにα1.2FT遺伝子を導入する。ES細胞で遺伝子導入後α1.2FT遺伝子導入された細胞はヒト血清による補体依存性細胞障害活性を受けないという特徴を用いてヒト血清によって選択する。選択された細胞はα1.3GTをノックアウトし、しかもα1.2FT遺伝子を導入された細胞ということになる。現在のベクターを用いて、我々はマウスES細胞に遺伝子導入を行い、ノックインされたマウスES細胞を選択している。今後これらの研究結果をもとに、さらに完成度の高いGALαGALの発現が抑制されたトランスジェニックブタを作成する予定である。
