Research Abstract	1.扁平上皮癌に関して (1)ヒト食道扁平上皮癌培養細胞(TE1とTE2)を、免疫細胞化学、ウェスタンプロット、ノーザンプロットで検索し、扁平上皮癌培養細胞に発現しているトロンボモジュリン(TM)と血管内皮細胞に存在するTMは同一であることを明らかにした。さらに抗凝固活性の測定により、扁平上皮癌細胞も抗凝固活性を有することを明らかにした。 (2)肺扁平上皮癌34例や口腔扁平上皮癌65例でも、原発巣とリンパ節転移巣におけるTMの発現を比較し、食道扁平上皮癌と同様、リンパ節転移巣でのTM発現が原発巣より有意に減少していることを明らかにした。 (3)ヒト食道扁平上皮癌の転移巣から得られた培養株(TE3)より、蛍光励起細胞分離装置(FACS)を用いて、TM高発現株とTM低発現株を分離し、双方の浸潤能の差をinvasion assay法によって検討したところ、TM低発現株はTM高発現株より明らかに浸潤能が高いことが判明し、扁平上皮癌転移巣でのTM発現の減少との関連が示唆された。 2.腺癌に関して (1)膵や肝内胆管の腺癌のin situ hybridizationによる検索で、浸潤傾向の弱い癌に特徴的なMUC2ムチンの発現は、MUC2mRNAにより制御されており、MUC2mRNAの発現が癌患者の予後良好因子になることを明らかにした。 (2)膵や肝内胆管の腺癌の組織切片でのムチンの発現様式のシアリダーゼ処理による変化から、浸潤傾向が強く予後不良の癌に高率に発現するMUC1ムチンはシアル酸エピトープを持つことが判明した。一方、浸潤傾向の強い癌と弱い癌との間のMUC2ムチンの染色性の違いにはシアル酸エピトープは関与しないことも明らかとなった。 (3)ファータ-乳頭部癌や胃癌においても、膵や肝内胆管の腺癌と同様に、MUC1ムチンの発現は予後不良因子であり、逆に、MUC2ムチンの発現は予後良好因子であることを明らかにした。 (4)大腸や胃粘膜におけるMUC2ムチンコア蛋白とムチン型糖鎖であるシアリルTnの細胞内局在を二重染色を用いた免疫電顕で同時に超微形態的に観察することに成功し、腫瘍性病変における詳細な検索への応用が期待される。
